Nef is a protein encoded by the HIV (Human Immunodeficiency Virus) genome. It is an accessory protein that plays a crucial role in the pathogenesis of HIV infection. Nef is known for its ability to modulate the host cell environment to favor viral replication and persistence.

Structure

Nef is a myristoylated protein, which means it undergoes a post-translational modification where a myristoyl group is added to its N-terminal glycine residue. This modification is essential for its membrane association and function. The protein is approximately 27 kDa in size and consists of multiple domains that mediate interactions with various host cell proteins.

Function

Nef has several functions that contribute to the progression of HIV infection:

  • Downregulation of CD4: Nef reduces the expression of CD4 receptors on the surface of infected cells. This prevents superinfection and helps in the evasion of the host immune response.
  • Modulation of MHC-I: Nef downregulates MHC class I molecules, which helps infected cells evade detection by cytotoxic T lymphocytes.
  • Enhancement of viral infectivity: Nef increases the infectivity of progeny virions, although the exact mechanism is not fully understood.
  • Alteration of T cell signaling: Nef interacts with various signaling molecules within T cells, altering their activation and survival.

Nef is considered a critical factor in the pathogenesis of HIV. It is involved in the early stages of infection and contributes to the establishment of a persistent infection. By modulating host cell pathways, Nef enhances viral replication and helps the virus evade the immune system.
