Lisinopril/amlodipine is a combination medication used in the management of hypertension (high blood pressure). It combines two active ingredients: lisinopril, an ACE inhibitor, and amlodipine, a calcium channel blocker. This combination is effective in lowering blood pressure by relaxing blood vessels and reducing the workload on the heart.

Pharmacology

Lisinopril

Lisinopril is an ACE inhibitor that works by blocking the conversion of angiotensin I to angiotensin II, a potent vasoconstrictor. This action leads to vasodilation, reduced blood pressure, and decreased aldosterone secretion, which helps in reducing sodium and water retention.

Amlodipine

Amlodipine is a calcium channel blocker that inhibits the influx of calcium ions into vascular smooth muscle and cardiac muscle. This results in vasodilation and a decrease in peripheral vascular resistance, which lowers blood pressure. Amlodipine is particularly effective in treating hypertension and angina pectoris.

Indications

Lisinopril/amlodipine is indicated for the treatment of hypertension in patients who require multiple drugs to achieve adequate blood pressure control. It may also be used in patients with chronic stable angina or vasospastic angina when monotherapy is insufficient.

Dosage and administration

The dosage of lisinopril/amlodipine is individualized based on the patient's response and tolerance. It is typically administered once daily, with or without food. The combination is available in various strengths to accommodate different therapeutic needs.

Side effects

Common side effects of lisinopril/amlodipine include dizziness, headache, edema, and cough. Serious side effects may include angioedema, hypotension, and renal impairment. Patients should be monitored for signs of allergic reactions and electrolyte imbalances.

Contraindications

Lisinopril/amlodipine is contraindicated in patients with a history of angioedema related to previous ACE inhibitor therapy, pregnancy, and in those with known hypersensitivity to either component of the combination.
